This is what happened when I washed a new to me backpack :

I put about a tablespoon of powder laundry detergent and some cold water in the wash basin and shook the backpack around. The water turned brown pretty quick.
Pretty sure it's just dirt from normal use, but who the heck knows... There were no visible stains, smells, or anything to indicate it was something gross. I suspect it was just dust and dirt that accumulated over several years of use either from hiking or some kid used it during high school where it sits on the floor of the classroom, ball field, transit bus, etc.
